Mammalian MAPs, MAP2, MAP4, and Tau, have a repeating site of the \"AP (assemblypromoting) sequence\" in the central part of the microtubule-associated site that directly binds to microtubules. Recently, genetic analysis revealed that molecules with repeated AP sequences exist throughout the animal kingdom, but it remains unclear whether these molecules function as MAPs. The purpose of this study was to identify the primordial MAP molecule using the marine planarian, Stylochoplana pusilla . First, we examined the breeding method to maintain S. pusilla in the laboratory. As a result of changing the diets, the S. pusilla chased and preyed on brine shrimp, and the body size and survival rate of the group that ate the brine shrimp were high. Furthermore, two S. pusilla MAP (SpMAP) molecules were identified based on RNA-seq data. The predicted amino acid sequences were analyzed based on the cloned sequences of SpMAP mRNA. As a result, the two types of SpMAPs had different numbers of AP sequence iterations, and there were SpMAP_4R with 4 repeating AP sequences and SpMAP_5R with 5 repeating AP sequences. SpMAP_4R was missing the 4th AP sequence of SpMAP_5R. Mammalian neural MAPs, MAP2 and τ, lack the second AP sequence of systemic MAP4, so SpMAPs may also show differences in localization and function such as neuronic and systemic.
