Although the morphology itself and process of stem development have been extensively investigated, it remains unknown how mechanical forces and stem organogenesis are coordinated. Good examples to advance our understanding of the mechanical properties of the stem organ are the recently identified mutant lines of Arabidopsis thaliana , which display spontaneous cracks in the stem. In these mutant lines, the morphologies at certain fixed time-points are becoming clearer. However, morphologies at the time-point when the stem begins to crack, and those at the time of its extending process are still unclear. Accordingly, in this research, we set up a method to acquire timelapse images and succeeded in capturing the process of stem cracking from the beginning to extending stages.
